El premio Grammy a la mejor interpretación de rock de un dúo o grupo con vocalista fue un galardón entregado en la ceremonia anual de los premios Grammy, una ceremonia establecida en 1958 originalmente llamada premios Gramophone, a los artistas que graban música —canciones o álbumes— dentro del género musical del rock. Los premios Grammy son una ceremonia anual presentada por The Recording Academy de los Estados Unidos para «distinguir a los logros artísticos, la pericia técnica y la excelencia en general en la industria de la grabación, sin tener en cuenta la cantidad de ventas del álbum o su posición en las listas».

The Recording Academy reconoció por primera vez a los artistas de rock en la 22.ª edición del los premios Grammy, cuando creó las categorías de mejor interpretación vocal de rock femenina, mejor interpretación vocal de rock masculina, mejor interpretación instrumental de rock y mejor interpretación rock de un dúo o grupo con vocalista. Según la descripción de cada categoría en la 52.ª edición, se otorga a «interpretaciones vocales grabadas recientemente por dúos, grupos o colaboraciones».

U2 posee el récord de la mayor cantidad de premios ganados, con un total de siete y Aerosmith por su parte, recibió cuatro. Entre los que lo han ganado dos veces se cuentan The Police y Kings of Leon. Green Day posee el récord de la mayor cantidad de nominaciones sin haber ganado, con cinco.

El premio dejó de otorgarse en 2011 debido a una reorganización general de las categorías de los premios Grammy. A partir del año siguiente, todas las interpretaciones de rock se ubicaron en la categoría de mejor interpretación rock.
